Item Beneficial effects of curcumin nano-emulsion on spermatogenesis and reproductive performance in male rats under protein deficient diet model: enhancement of sperm motility, conservancy of testicular tissue integrity, cell energy and seminal plasma amino acids content(BioMed Central, 2017-12) Ahmed-Farid, Omar AH; Nasr, Maha; Ahmed, Rania F; Bakeer, Rofanda MMalnutrition resulting from protein and calorie deficiency continues to be a major concern worldwide especially in developing countries. Specific deficiencies in the protein intake can adversely influence reproductive performance. The present study aimed to evaluate the effects of curcumin and curcumin nano-emulsion on protein deficient diet (PDD)-induced testicular atrophy, troubled spermatogenesis and decreased reproductive performance in male rats. Juvenile rats were fed the protein deficient diet (PDD) for 75 days. Starting from day 60 the rats were divided into 4 groups and given the corresponding treatments for the last 15 days orally and daily as follows: 1st group; curcumin group (C) received 50 mg/kg curcumin p.o. 2ndgroup; curcumin nano-form low dose group (NCL) received 2.5 mg/kg nano-curcumin. 3rd group; curcumin nano-form high dose group (NCH) received 5 mg/kg nano-curcumin. 4th group served as malnutrition group (PDD group) receiving the protein deficient diet daily for 75 days and received distilled water ingestions (5 ml/kg p.o) daily for the last 15 days of the experiment. A normal control group was kept under the same conditions for the whole experiment and received normal diet according to nutrition requirement center daily for 75 days and received distilled water ingestions (5 ml/kg p.o) daily for the last 15 days of the experiment. Steroid hormones such as estrogen and progesterone are thought to play an important role in the process of carcinogenesis of ovarian tumors. There are two subtypes of the nuclear estrogen receptor (ER-α and ER-β) encoded by separate genes. This work aimed to evaluate the expression pattern of ER-α and ER-β in epithelial ovarian carcinoma and their correlation with tumor histopathological parameters and proliferating cell nuclear antigen expression as a proliferation marker. Materials and methods A total of 50 cases of epithelial ovarian carcinoma were included in this study. All cases were female patients who underwent oophorectomies or subtotal or total hysterectomies with oophorectomies. Surgical specimens were sent to Pathology Department at Kasr El-Aini hospitals and to private laboratories. The cases were graded and staged according to WHO systems. The cases were stained by hemotoxylin and eosin for histopathological grading, and they were immunohistochemically stained for ER-α, ER-β, and proliferating cell nuclear antigen using streptavidin–biotin technique. Results In this study, 56% of cases were positively stained for ER-α. It is significantly correlated with both of the tumor histological type and proliferative state of the tumors. There was a significant inverse correlation between ER-α expression and the tumor histological grade. Approximately 62% of cases were positively stained for ER-β. There was a significant inverse correlation between ER-β positivity and both of the tumor stage and proliferative state of ovarian carcinoma cases. Conclusion The loss of ER-β, not ER-α, expression in ovarian tumors may be a feature of malignant transformation suggesting its potential role as tumor suppressor gene. Determination of ER subtypes may improve response to hormonal therapy using a selective ER modulator in selected cases of ovarian carcinoma.Item Hepatoprotective effect of Omega-3 PUFAs against acute paracetamol- induced hepatic injury confirmed by FTIR(SAGE, 2020) El-Gendy, Zeinab A; El-Batran, Seham A; Youssef, SAH; Ramadan, A; El Hotaby, Walid; Bakeer, Rofanda MAcute paracetamol over dose-induced hepatotoxicity is considered an important medical hazard especially among women. Omega-3 long-chain polyunsaturated fatty acids (Omega-3 PUFAs) daily doses are nowadays recommended for their antioxidant and anti-inflammatory potentials. Fourier transform infrared (FTIR) spec- troscopy is considered a reliable method in analyzing cellular alterations and is now efficiently used to diagnose several diseases and the efficacy of drugs even in the early stages. The aim of our study was to evaluate the hepatoprotective effect of Omega-3 PUFAs against paracetamol-induced hepatotoxicity in rats confirmed through measuring protein alterations in hepatocytes by FTIR. Rats were pretreated with Omega-3 PUFAs (50 and 100 mg/kg) for 21 days prior to oral ingestion of paracetamol. FTIR results revealed that Omega-3 PUFAs (50 mg/kg) limited the toxic effects of paracetamol by restoring the hepatic amide I to amide II ratio. In addition; biochemical analyses demonstrated that serum ALT, AST, Cholesterol, LDL-cholesterol and Il-6 levels as well as hepatic TNF-a, MDA, NOx levels were decreased. Besides; serum HDL-cholesterol level and hepatic GSH level were increased. Histopathological examinations of hepatic sections validated the hepatoprotective potential. The overall effect of this dose was comparable to those of the usual recommended hepatoprotective supplement; silymarin. LC–MS/MS together with molecular networking was used to profle the metabolites of LSLE. LSLE treated group revealed signifcant changes in alopecia related biomarkers, perturbation of androgenic markers; decline in testosterone level and elevation in 5α-reductase (5-AR); decline in the cholesterol level. On the other hand, LSLE treated group showed improvement in vascular markers; CTGF, FGF and VEGF. Groups treated topically with minoxidil and LSLE showed signifcant improvement in hair length. LC–MS/MS profle of LSLE tentatively identifed 17 constituents: mainly glucosinolates, favonoid glycosides, alkaloids and phenolic acids. The results point to the potential role of LSLE in the treatment of alopecia through decreasing 5(alpha)- dihydrotestosterone levels. Molecular docking was attempted to evaluate the probable binding mode of identifed compounds to androgen receptor (PDB code: 4K7A).
