Item Antihyperglycemic and antihyperlipidemic effects of the methanol extracts of Cleome ramosissima Parl., Barleria bispinosa (Forssk.) Ghareib, SalahThe antihyperglycemic and antihyperlipidemic effects of the methanolic extracts of the aerial parts of Cleome ramosissima Parl. (Cleomaceae), Barleria bispinosa (Forssk.) Vahl. (Acanthaceae) and Tribulus macropterus Boiss. (Zygophyllaceae) were evaluated in streptozotocin (STZ) induced diabetic rats at a dose of 500 mg/kg bw. The reduction in fasting blood glucose level (BGL) was observed in the following order C. ramosissima, B. bispinosa and T. macropterus at the 4th week of administration. C. ramosissima and T. macropterus also showed significant increase in plasma insulin by 100.6% and 189.9%, respectively. The studied plant extracts induced an increase in both utilization and tolerance of glucose in diabetic rats. The hypolipidemic effect of C. ramosissima and T. macropterus was demonstrated by a significant reduction in plasma total cholesterol (TC) (42.6% and 37.2%, respectively) and low density lipoprotein cholesterol (LDL-C) (48.0% and 42.1%, respectively) and the increase of high density lipoprotein cholesterol (HDL-C) by 81.0% and 91.9%, respectively. B. bispinosa decreased the blood levels of LDL-CItem Antiprotozoal activity of major constituents from the bioactive fraction of Verbesina encelioides(Taylor & Francis, 2017) Ezzat, Shahira M; M Salama, Maha; A Mahrous, Engy; Maes, Louis; Pan, Cheol-Ho; Abdel-Sattar, EssamThe bioactive petroleum ether fraction of Verbesina encelioides, previously studied by the authors, was chosen for the isolation of antiprotozoal metabolites. Pseudotaraxasterol-3β-acetate (1), benzyl 2,6-dimethoxy benzoate (2), 16β-hydroxy-pseudotaraxasterol-3β-palmitate (3) and pseudotaraxasterol (4), in addition to β-sitosterol glucoside (5) and β-sitosterol galactoside (6) were isolated and identified based on one-dimensional and two-dimensional spectral analysis. This is the first report describing (3) and (6) in genus Verbesina. The isolated compounds were tested in vitro against Plasmodium falciparum, Trypanosoma brucei, Trypanosoma cruzi and Leishmania infantum. Cytotoxicity was evaluated on MRC-5 cells. Compound 1 showed moderate to weak activity against L. infantum T. brucei and P. falciparum and was inactive against T. cruzi. Compound 3 showed moderate activity against L. infantum, compound 4 revealed weak activity against T. cruzi, while 5 and 6 were inactive against all tested protozoa. All compounds were non-cytotoxic. The isolated constituents showed less antiprotozoal activity than the crude fraction.Item Chemical and biological assessment of high value pharmaceutical raw material from Egyptian orange peel(National Institute of Science Communication, 2022-06) Meselhy, Meselhy R; Ezzat, Shahira M; Ali, Dalia El-Sheikh; El-Adl, Mohamed; El-Shafei, Reham A; Abd El-Aziz, Marwa M; Abdel-Sattar, EssamIn Egypt, orange juice industries generate huge amount of waste peel that could be a source of raw materials with high value and economic return. Here, we explored a better model for waste management of citrus processing waste in Egypt by developing an ecofriendly method for preparation of value added materials. A high grade pectin (HGP) was obtained from the crude acidic water extract of the peel after passing through Diaion HP20 column. The HGP showed potent antidiabetic activity at lower dose than those reported in literature. It possessed significant effect on blood glucose (BG) level, as well as parameters relevant to liver and kidney functions in streptozotocin (STZ) induced diabetic rats. On the other hand, the essential oil (EO) prepared by cold press showed the highest yield (0.72% w/w) and it is the most applicable method of isolate orange oil on pilot scale. EO showed significant antimicrobial activity against the tested food borne pathogens. In conclusion, high value materials — HGP and EO were prepared on pilot scale from the waste orange peel. While the HGP can be included in food supplement for diabetic patients, EO can be used as a natural food preservative.Item Chemical and Biological Investigation of Araucaria heterophylla Salisb. Mouneir, SamarThree labdane diterpenes, namely labda-8(17),14-diene, 13-epicupressic acid, and 13-Oacetyl- 13-epicupressic acid, were isolated from the resin collected from stem exudates of Araucaria heterophylla Salisb. (Araucariaceae). The isolated compounds were identifi ed using different spectroscopic methods (1H NMR, 13C NMR, HMQC, HMBC and COSY). The resin extract showed antiulcerogenic activity against ethanol-induced stomach ulcers in Sprauge Dawely rats using ranitidine as standard. In addition, the resin and the isolated compounds showed variable cytotoxic activities against breast (MCF7) and colon (HCT116) cancer cell lines.Item Metabolomic profiling of barley extracts obtained via different solvents and evaluation of their anti-inflammatory efficacy(Elsevier B.V, 2024-07) Eid, Omneya; Salem, Mohamed A; Mohamed, Osama G; Ezzat, Shahira; Tripathi, Ashootosh; Ewida, Menna A; El Sayed, Abeer; Abdel-Sattar, Essam; Elkady, Wafaa MBarley (Hordeum vulgare L.) stands out for its special nutritional value compared to other cereals, garnering significant attention in recent years. In this study, a comprehensive chemical and biological evaluation of different barley cultivars extracted with different solvents was performed. Ultra-high performance liquid chromatography coupled with electrospray ionization quadrupole time-of-flight tandem mass spectrometry (UHPLC-ESI-QTOF-MS/MS) was used in the phytochemical study. The molecular docking with absorption, distribution, metabolism, excretion, and toxicity (ADMET) study and multivariate analysis alongside the quantification of total phenolic contents (TPC) were performed. Furthermore, the anti-inflammatory action was in vitro assessed by measuring COX-1, COX-2, and 5-LOX inhibition. This study seeks to establish a suitable solvent for extracting anti-inflammatory properties from diverse barley cultivars. The methanol extract obtained from the G.131 cultivar, followed by the 80% methanol extract from the G.136 cultivar, exhibited the most favorable IC50 values in comparison to the reference drugs indomethacin and celecoxib. The phytochemical profile revealed more than 60 compounds that belonged to nitrogen-containing compounds, phenolic compounds, organic acids, flavonoids, and their glycosides, hordatines, and other classes. The TPC and COX-2 inhibition was positively correlated with isoscoparin, coumaroyle–OH–eagmatine, isoorientin, isovitexin 7-O-rhamnosylglucoside, isoscoparin 7-O-glucoside, and isoorientin-7-O-[6-feruloyl]-glucoside. These findings along with their pharmacodynamics study suggested a potential synergistic and significant contribution of these compounds with the anti-inflammatory activity.Item Protective effect of Echinops galalensis against CCl4-induced injury on the human hepatoma cell line (Huh7)(Elsevier, 2013) Ezzat, Shahira M; M. Abdel-Naim, AshrafPhytochemical investigation of the flowering aerial parts of Echinops galalensis (Asteraceae) led to the isolation of a new taraxasteryl triterpene, 3b-acetoxy-taraxast-12, 20(30)-diene-11a-21a-diol (1), together with nine known metabolites, a-amyrin (2), b-sitosterol (3), erythrodiol (4), lup-20(29)-ene- 1,3-diol (5), 1,5-dicaffeoylquinic acid (6), 3,5-dicaffeoylquinic acid (7), 3,4-dicaffeoylquinic acid (8), 4,5- dicaffeoylquinic acid (9) and apigenin-7-O-b-D-glucoside (10). The structure of the new compound was determined by comprehensive analyses of their 1D and 2D NMR, mass spectral (HR-EI) data and comparison with previously known analogs. The effect of the methanol extract of E. galalensis, its fractions as well as compounds (1–10) on human hepatoma cell line (Huh7) was evaluated according to aspartate aminotransferase (AST), alanine transaminase (ALT), superoxide dismutase (SOD) activities and malondialdehyde (MDA) level before and after exposure of the cells to carbon tetrachloride (CCl4). It was found that pre-treatment of human hepatoma cell line (Huh7) with the tested samples (100 mg/ml) prior to CCl4 challenge protected against cell injury. The protective effect of E. galalensis was suggested to be mediated, at least partly, by its antioxidant activity.Item Pumpkin Bio-Wastes as Source of Functional Ingredients(Springer, 01/01/2022) Ezzat, Shahira M; Adel, Riham; Abdel-Sattar, EssamFood processing wastes are those end products of various food processing industries that have not been recycled or used for other purposes. They are the non-product flows of raw materials whose economic values are less than the cost of collection and recovery for reuse; therefore discarded as wastes. These wastes could be considered valuable by-products if there were appropriate technical means and if the following products’ value exceeded the reprocessing cost. A tremendous amount of food wastes is generated worldwide, largely through the industry of fruit and vegetable, oils, fermentation, dairy products, and seafood. Wastes are generated at the different food processing stages: pre- and post-harvest, food manufacturing, packaging, retail/catering, and consumer/household. The loss can reach up to 25% or more worldwide (Parfitt et al., 2010). In the developed leading countries, such as Japan, the USA, and United Kingdom, more than 40% was discarded as food waste.Item Valorization of Egyptian Food Byproducts in the Development of Biologically Active Nutraceuticals.(IJP, 2018) Abdel-Sattar, Essam; Amer, Reham I.; Bagrel, Denyse; El Mansourai, Latifa; Philippot, Stéphanie; Darwish El-Tanbouly, Nebal; Evain-Bana, Emilie; Boukhira, Smahane; Abou-Hussein, Dina R.; Bousta, Dalila; Tzanova, Tzvetomira; Salah El Dine, RihamThis study was aimed to maximize the benefits of the use of by-products (oil cake) of olive fruits and black seeds after extraction of their fixed oils, rather than their use as animals feed or landfilling or composting. The cake of both seeds was assessed by HPLC for their main bioactive marker compounds (thymoquinone for black seed and oleuropein for olive fruit), to choose the best method for extraction, determination of phenolic contents and in vitro antioxidant activity (DPPH and reducing powers, FRAP assays). Both by-products were also assessed for their cytotoxicity against four human breast cell lines, three of them are cancerous (MCF7, MDA-MB-213, Vcr-R), and a non-cancerous one (epithelial type) but immortalized by telomerase (hTERT-HME), in addition to a human hepatoma cell line (HepG2). Also, both wastes were subjected for in-vitro CDC25s phosphatase inhibition assay on three isoforms (A-C) and for in-vivo immunomodulatory effects. In conclusion, the results of this study showed the interest of cumulating different biological approaches exploring various physiological mechanisms and showed the utility of these extracts in different fields, the first being used as a cytoprotective agent, and the second one being promising as an anticancer agent.
